Calcein staining of calcified structures in pearl oyster Pinctada margaritifera and the effect of food resource level on shell growth ArchiMer

Marine mollusc shell growth has been widely measured using fluorochrome marking. In order to test the efficiency and reliability of calcein staining on P. margaritifera shells and pearls, the present study examined two administration methods, different concentrations and several immersion times.Immersion in a 150 mg.L-1 calcein solution for 12 h to 24 h appeared to be the best method for marking P. margaritifera shells. For pearl marking, injection of a 200 mg.L-1 calcein solution into the pearl pouch was the optimal method. Calcification abnormalities linked to the appearance of a brown coloration of the internal shell layer are observed in scallops Pecten maximus in the Brest roadsted. It is a widespread phenomenon in shallow waters (about 3 to 10 meters) sometimes associated with an important boring parasitism at the ultimate stage of development. This brown color which can generate a decrease of the consumer appraisal does not affect the commercial value of the shellfish (muscle and gonad quality) when it is not too high. Structural and biochemical characteristics in adult scallop shells ArchiMer

This paper presents results of studies carried out on the microstructure, composition of organic matter and amino acids in abnormal shells of adult Pecten maximus. Results indicate that abnormal P. maximus shells can have two types of brown material on the inner layer of the valves. Brown calcified areas can be present and they have abnormal crystals on the internal foliated layer of the shell. An index (MIBC) was developed to describe this type of abnormality that depended on the extent of the brown areas. The other type abnormality had brown material consisting of uncalcified membranes that were embedded in mud tubes made by Polydora sp. worms.
